Studies on the enzymatic formation of citrovorum factor by Streptococcus faecalis.

چکیده
There is evidence that before pteroylglutamic acid (PGA) can carry out certain catalytic functions in animal tissues it must first be converted to citrovorum factor (CF). This conversion has been studied in rat liver slices by Nichol and Welch (l), who found that the conversion was enhanced by ascorbic acid. Since Streptococcus faecalis utilizes either PGA or CF to meet its growth requirement for folic acid (2), this organism presumably carries out this conversion efficiently. This organism might therefore be conveniently employed to study the enzymatic transformation of PGA to CF by the resting cell technique. The biochemical conditions favoring this transformation by S. faecalis and by a strain of X. faecalis resistant to 4-amino-lo-methylpteroylglutamic acid (Amethopterin) are the subject of this report.

Pyrimethamine (Daraprim (Burroughs Wellcome) ; 2,4diamino-5-p-chlorophenyl-6-ethylpyrimidine) is used in the treatment and prevention of malaria (1). When used in combination with one of the sulfonamides, it is also effective in the treatment of toxoplasmosis (2) and coccidiosis (3). Pyrimethamine is one of a series of 2,4-diaminopyrimidines which have been shown to be antagonists of folic acid...

The discovery of the citrovorum factor (CF) in yeast and in liver (Sauberlich and Baumann, 1948) was accompanied by the observation that a portion of the material existed in a bound form, from which it was liberated by incubation with an enzyme system ("conjugase") found in hog kidney (Bird et al., 1946). Bound forms of CF were found by Iwai et al. (1954) in higher plants, and these also contai...

May and coworkers (l-3) demonstrated that the production of a scorbutic state in monkeys led to the development of a megaloblastic anemia, a condition which responded promptly to treatment with pteroylglutamic acid (PGA) or to very much smaller amounts of synthetic citrovorum factor (leucovorin or folinic acid-SF) .I It has been shown that the administration of ascorbic acid to normal human sub...

Sauberlich and Baumann (1948) have reported that Leuconostoc citrovorum, strain 8081, failed to grow on a synthetic medium unless the medium was supplemented with an unidentified growth factor isolated from liver extracts, designated citrovorum factor. More recent reports in the literature (Sauberlich, 1949; Nichol and Welch, 1950a) point to the conversion of pteroylglutamic acid to citrovorum ...

1. The effects of dietary biotin compared with vitamin B(12) on the total content and on the distribution of the various folate derivatives in the liver of rats given a biotin-free diet have been studied. The effect of both vitamins on the conversion in vitro of folic acid into citrovorum factor in the same experimental conditions was also examined. 2. In biotin-treated rats as well as in vitam...
